How Is OvoPro Made? A Research Review of Precision-Fermented Egg-White Protein

OvoPro is not a plant-based egg substitute and it is not a complete egg grown outside a hen. The EVERY Company describes it as a powdered, precision-fermented ovalbumin ingredient—the major protein in chicken egg white.
The closest public U.S. regulatory record is FDA GRAS Notice 1104 from Clara Foods Co., doing business as The EVERY Company. It describes an egg-white protein preparation made with engineered Komagataella phaffii yeast and consisting predominantly of recombinant ovalbumin. The public FDA documents do not use the OvoPro brand name, so this review treats GRN 1104 as the closest public regulatory dossier for EVERY’s ovalbumin ingredient rather than proof that every current commercial lot is identical to the material assessed in 2023.

Why This Review Matters
Ovalbumin accounts for about 54% of egg-white protein. Its amino-acid sequence, folding, heat response, surface behavior, and ability to form networks contribute to egg white’s foaming, binding, and gelling functions. Precision fermentation attempts to make this one protein without raising a hen or forming an egg.
This is a review of an ingredient-production system, not a poultry feeding trial. No hens, chicks, or human volunteers were enrolled. The evidence combines company-generated analytical studies submitted to FDA, FDA’s regulatory response, and peer-reviewed studies of recombinant ovalbumin produced in fungal hosts.
How OvoPro-Type Ovalbumin Is Created
Select the protein target. The company used a synthesized coding sequence for mature chicken ovalbumin, also called Gal d 2. Native ovalbumin contains 385 amino acids.
Build a secretion cassette. The ovalbumin sequence was joined to a Saccharomyces cerevisiae alpha-mating-factor secretion signal. This directs K. phaffii to process and export the new protein outside the yeast cell, simplifying recovery.
Engineer the yeast chromosome. Linear DNA cassettes containing a methanol-inducible promoter, the secretion signal, the ovalbumin sequence, and an AOX1 terminator were introduced by electroporation and integrated into chromosomal DNA. Additional HAC1 copies were added because HAC1 activates the unfolded-protein response and can improve folding and secretion of a foreign protein.
Select and stabilize the production strain. PCR and protein-expression assays identified DFB-004. The dossier reports that the introduced changes remained present after 45 generations without selection, selectable antibiotic markers were removed, and the production organism contains no antibiotic-resistance genes.
Run controlled fermentation. Cryopreserved yeast seed is expanded to a target cell density and transferred to a production bioreactor. The disclosed system grows biomass on glycerol and glucose, then uses methanol-responsive promoters to induce ovalbumin production. Culture purity is monitored during the run.
Recover the secreted protein. Centrifugation and microfiltration remove yeast cells from the fermentation liquid. Ultrafiltration concentrates the protein fraction. Column chromatography and pH adjustment further purify it, and spray drying produces the powder.
Verify identity and release quality. SDS-PAGE monitors protein identity during production. LC-MS/MS identifies ovalbumin and remaining host proteins. Final testing covers composition, heavy metals, and microorganisms.
What Is Actually Created?
The ingredient is predominantly recombinant Gal d 2, not a complete egg white and not a whole egg. Native egg white also contains ovotransferrin, ovomucoid, lysozyme, ovomucin, and many other proteins. An ovalbumin ingredient may replace selected egg functions without reproducing every property of a whole egg.
The public dossier reports a 389-amino-acid recombinant protein: the 385-amino-acid mature hen sequence plus a four-amino-acid EAEA extension remaining after secretion processing. It reports a calculated molecular mass of 43.15 kDa compared with 42.88 kDa for native ovalbumin. The recombinant protein also has different glycosylation. These are genuine biochemical differences even though the core chicken sequence is retained.
Key Findings
• Engineered yeast acts as the protein factory. Chicken cells, incubated embryos, and egg contents are not the production material.
• The yeast secretes ovalbumin into the broth; physical separation, filtration, and chromatography remove cells and concentrate the protein.
• The recombinant protein is highly similar but not molecularly identical to native ovalbumin.
• Three company-tested lots showed 99.3% to 99.7% in-vitro pepsin digestibility, compared with 99.3% for native Gal d 2 under the same method. This is a laboratory assay, not a human clinical trial.
• Thermal-shift testing reported melting temperatures of 72.6°C to 73.2°C for three recombinant lots versus 74.4°C for native Gal d 2.
• The egg allergen is preserved. FDA’s response states that the protein will likely trigger reactions in egg-allergic consumers and that U.S. egg-allergen labeling applies.
• FDA’s October 17, 2023 letter says the agency had no questions regarding the notifier’s GRAS conclusion for stated uses. The letter expressly says this is not an affirmation under 21 CFR 170.35.
Strengths of the Evidence
The FDA dossier makes the production logic, strain lineage, purification train, product specifications, and analytical comparisons publicly inspectable. FDA requested clarifications and amendments involving sequence length, thermostability, purification, dietary exposure, and heavy-metal specifications. Three nonconsecutive lots were evaluated against release specifications. Several methods—including DNA sequencing, PCR, SDS-PAGE, LC-MS/MS, digestibility testing, and thermal analysis—were used to distinguish protein identity from process purity.
Independent peer-reviewed studies support the broader principle that fungal hosts can express folded ovalbumin with useful foaming and gelling behavior. They also document structural and functional differences from hen-derived protein.
Limits of the Evidence
Most publicly available OvoPro-specific analytical and performance data were generated or sponsored by the manufacturer. GRN 1104 is a regulatory dossier, not a peer-reviewed clinical or food-functionality paper.
The FDA record names EVERY and its non-animal egg-white protein but not the OvoPro trademark. Current manufacturing partners may use updated scales, equipment, or controls not described in the 2023 file.
Commercial details such as full media composition, exact feed schedules, fermentation yield, economics, chromatography conditions, and current certificate-of-analysis ranges remain confidential or undisclosed.
Peer-reviewed Pichia and Trichoderma studies used different research organisms or strains. They support the mechanism and known challenges but do not independently validate current commercial OvoPro lots.
Company claims about binding, gelling, foaming, whipping, shelf life, cost savings, flavor, and protein quality are commercially relevant but are not all supported by full, independently peer-reviewed OvoPro datasets in the public record.
GRAS status addresses safety under specified intended uses. It does not prove that OvoPro is nutritionally or functionally superior to shell eggs, can replace yolk functions, or performs identically in every recipe. The evidence also does not measure poultry-production, farm-economic, laying-hen-welfare, avian-influenza, or environmental outcomes.
Poultry Nerds Takeaway
OvoPro is best understood as a purified egg-white protein made by a different biological factory. A hen normally expresses ovalbumin in the oviduct and deposits it into egg white. Precision fermentation moves the protein-making instruction into K. phaffii yeast, directs the yeast to secrete ovalbumin into fermentation broth, and then separates and purifies that protein into powder.
The science supports the core manufacturing claim: engineered yeast can produce a highly similar ovalbumin that retains major structural and functional features. Careful wording still matters. The recombinant ingredient differs in its N-terminal sequence and glycosylation, contains one principal egg protein rather than the complete egg-white system, and remains an egg allergen.
For poultry keepers and educators, this is a noteworthy food-technology development—not evidence that conventional eggs have become obsolete. Whether the ingredient is economical, environmentally preferable, or functionally interchangeable must be demonstrated application by application.
